Biological Background: CD83 Function and Localization
- CD83 is a transmembrane glycoprotein predominantly expressed on the surface of various immune cells, including dendritic cells and lymphocytes.
- It plays an essential role in CD4+ T-cell selection, differentiation, and stability by regulating the activity of the E3 ubiquitin ligase MARCHF8, which controls MHCII trafficking.
- CD83 inhibits MARCHF1-mediated ubiquitination of MHCII and CD86, preventing their degradation and supporting antigen presentation (PubMed:21220452). Related references: PMID:21220452
- It acts as an important modulator of protective responses against acute infections.
- The protein is localized to the cell membrane and contains an immunoglobulin domain, disulfide bonds, and glycosylation sites.
- CD83 is expressed by activated lymphocytes, Langerhans cells, and activated dendritic cells, reflecting its role in immune activation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1-100); consider validating specificity in lysates or fixed cells from activated immune cells.
- For western blot (WB), use appropriate loading controls and validate band size (~23 kDa) in relevant species (human, mouse, rat).
- In immunofluorescence (IF-P), optimize fixation and permeabilization conditions for membrane protein detection.
- For ELISA, consider pairing with a detection antibody that recognizes a distinct epitope to avoid competition.
