Biological Background: CD83 Function and Localization
- CD83 is a transmembrane glycoprotein predominantly expressed on immune cells such as dendritic cells and lymphocytes. It is also known as B-cell activation protein and cell surface protein HB15.
- It plays an essential role in CD4+ T-cell selection, differentiation, and stability by regulating the activity of MARCHF8, the major E3 ubiquitin ligase controlling MHCII trafficking. Related references: PMID:21220452
- CD83 inhibits MARCHF1 association with MHCII or CD86, preventing their ubiquitination and subsequent degradation. Related references: PMID:21220452
- It acts as an important modulator of protective responses against acute infections.
- Subcellular localization: CD83 is a membrane protein.
- It is specifically expressed in activated lymphocytes, Langerhans cells, and activated dendritic cells.
- The protein features a signal peptide, an immunoglobulin domain, disulfide bonds, and a transmembrane helix. It is a glycoprotein identified by direct protein sequencing and proteomics.
- The predicted molecular weight of CD83 is approximately 23 kDa (23042 Da), consistent with its glycosylated transmembrane nature.
Experimental Guidance and Technical Tips
- The antibody was generated against a recombinant protein spanning the extracellular domain (amino acids 20-143) of human CD83, suggesting it may recognize the native protein in membrane fractions or whole cell lysates.
- For Western blot, anticipate a band around 23 kDa; consider optimizing lysis and reducing conditions.
- For ELISA, this antibody may serve as a capture or detection reagent; validate performance in your specific sample system.
- As a rabbit monoclonal antibody, it is expected to offer high specificity and lot-to-lot consistency; confirm with appropriate controls.
