Biological Background: CD82 Function and Localization
- CD82 (KAI1, TSPAN27) is a metastasis suppressor and member of the tetraspanin family, also known as C33, IA4, R2, SAR2, ST6, 4F9, GR15.
- It acts as a structural component of tetraspanin-enriched microdomains (TERMs), platforms for receptor clustering and signaling (PubMed:19497983). Related references: PMID:19497983
- CD82 attenuates EGF signaling by facilitating ligand-induced EGFR endocytosis and desensitization via PKC-mediated CBL phosphorylation (PubMed:10985391, PubMed:35538033, PubMed:23897813). Related references: PMID:10985391, PMID:35538033, PMID:23897813
- It regulates cell-matrix adhesion and migration by modulating integrins ITGA4, ITGA6, and ITGB1 organization (PubMed:24623721, PubMed:8757325, PubMed:15557282, PubMed:17560548). Related references: PMID:24623721, PMID:8757325, PMID:15557282, PMID:17560548
- CD82 decreases plasminogen activation by interfering with PLAU-PLAUR interaction (PubMed:15677461). Related references: PMID:15677461
- CD82 delivers costimulatory signals for the TCR/CD3 pathway, controls TLR9 trafficking and myddosome assembly, and promotes anti-inflammatory macrophage polarization. It also inhibits LPS-induced inflammatory signaling by preventing LPS binding to TLR4 (PubMed:36945827). Related references: PMID:36945827
- CD82 suppresses angiogenesis by sequestering VEGFA and PDGFB, inhibiting their receptor signaling (PubMed:34530889). Related references: PMID:34530889
- Subcellularly, CD82 localizes to the cell membrane, cytoplasmic vesicles, and phagosomes; it is lymphoid-specific and subject to palmitoylation, glycosylation, and alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen spans the large extracellular loop (aa 168–267) of CD82, a region expected to be exposed in both native and denatured conditions, supporting use in Western blot (predicted molecular weight ~30 kDa, but glycosylation may shift observed band to 30–55 kDa) and ELISA.
- For immunofluorescence applications (IF/ICC, IF-P) and IHC-P, optimize fixation/permeabilization conditions; antigen retrieval may be necessary for paraffin-embedded tissues.
- Cross-reactivity with mouse CD82 allows use in mouse models, but validate expression and epitope conservation in the specific strain/tissue.
- Confirm antibody specificity using positive and negative controls (e.g., CD82-overexpressing cells or CD82 knockout samples) and titrate for each application.
