Biological Background: CD82 Function and Localization

  • CD82, also known as KAI1, TSPAN27, and several other aliases, is a member of the tetraspanin superfamily of cell surface glycoproteins, characterized by four transmembrane domains and conserved functional regions.
  • It serves as a structural component of tetraspanin-enriched microdomains (TERMs) that organize receptor clustering and signaling, regulating cell adhesion, migration, and protein trafficking. Related references: PMID:19497983
  • CD82 functions as a metastasis suppressor, inhibiting cell migration by modulating integrin-mediated adhesion through integrins such as alpha4/ITA4, alpha6/ITGA6, and beta1/ITGB1, thereby increasing cell-matrix adhesion. Related references: PMID:24623721, PMID:8757325, PMID:15557282, PMID:17560548
  • It attenuates EGF signaling by facilitating ligand-induced endocytosis and ubiquitination of EGFR, leading to receptor desensitization. Related references: PMID:10985391, PMID:35538033, PMID:23897813
  • CD82 decreases cell-associated plasminogen activation by interfering with the interaction between urokinase-type plasminogen activator (PLAU) and its receptor PLAUR. Related references: PMID:15677461
  • In immune cells, CD82 associates with CD4 or CD8 to provide costimulatory signals for the TCR/CD3 pathway and is involved in TLR9 trafficking to acidified compartments for myddosome assembly (By similarity). It also inhibits LPS-induced inflammation by blocking LPS binding to TLR4. Related references: PMID:36945827
  • CD82 controls angiogenesis by binding and sequestering VEGFA and PDGFB, inhibiting their signaling and promoting vascular quiescence. Related references: PMID:34530889
  • The protein is localized to the cell membrane, cytoplasmic vesicles, and phagosomes, where it participates in antigen presentation. It is palmitoylated and glycosylated, with a molecular weight of ~30 kDa (predicted 29.6 kDa), and its expression is lymphoid-specific.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 150–250 of human CD82 (intracellular region). For Western blot (WB), a specific band near 30 kDa is expected; consider using reducing conditions and validate with positive controls.
  • For immunohistochemistry (IHC-P), antigen retrieval may be necessary to expose the intracellular epitope; optimize fixation and retrieval protocols for your tissue type.
  • In immunofluorescence/immunocytochemistry (IF/ICC), permeabilization is required to access the intracellular epitope; include appropriate negative controls (e.g., peptide blocking) to confirm specificity.
  • This antibody cross-reacts with mouse and rat CD82, but due to sequence variation, verify epitope conservation before use in these species; pilot experiments are recommended.
  • For ELISA, the antibody can be used in direct or indirect formats; optimal coating and detection conditions should be determined empirically.
