Biological Background: CD80 Function and Localization
-
CD80 (B7-1, BB1) is a T-lymphocyte activation antigen belonging to the immunoglobulin superfamily, playing a pivotal role as a costimulatory molecule in T-cell activation (PubMed:38467718).

PMID:38467718 -
It serves as a ligand for the stimulatory CD28 receptor on naive T cells, providing the essential secondary signal for activation, and also binds the inhibitory CTLA-4 receptor, which acts as a decoy to block CD28-mediated priming (PubMed:12196291, PubMed:10583602, PubMed:11279502).

PMID:12196291, PMID:10583602, PMID:11279502 -
Downstream signaling through CD28/CD80 activates NF-kappa-B and MAPK pathways, leading to cytokine production, and additionally stimulates glucose metabolism and ATP synthesis via the PI3K/Akt pathway (PubMed:10438913, PubMed:12121659).

PMID:10438913, PMID:12121659 -
CD80 also modulates PD-L1/PD-1 interactions to limit excessive inhibitory signals and plays a critical role in germinal center reactions by mediating B-cell–T-cell crosstalk for effective humoral immunity (PubMed:36727298) (By similarity).

This transmembrane glycoprotein is localized on the cell membrane and expressed primarily on antigen-presenting cells, including B cells, dendritic cells, and macrophages.
-
Post-translational modifications include glycosylation, phosphorylation, and palmitoylation; the protein features a signal peptide, an immunoglobulin V-type domain, and a transmembrane helix.
-
Additionally, CD80 can serve as a host cell receptor for adenovirus subgroup B during infection.
-
Multiple isoforms are generated through alternative splicing, contributing to functional diversity.
Experimental Guidance and Technical Tips
- For Western Blot, CD80 may appear as multiple bands ranging from 18 to 33 kDa due to post-translational modifications such as glycosylation; consider running both reducing and non-reducing gels to resolve the specific band.
- In Flow Cytometry, since CD80 is a cell surface protein, we recommend staining live, unfixed cells; titrate the antibody to determine the optimal concentration for your experimental system.
- For ELISA applications, the antibody can be utilized as either a capture or detection reagent; validate its performance with your specific antigen and buffer conditions.
- As this is a recombinant rabbit monoclonal antibody, it demonstrates high batch-to-batch consistency and is ideal for reproducible quantitative assays.
