Biological Background: CD74 Function and Localization
- CD74, also known as HLA class II histocompatibility antigen gamma chain or invariant chain (Ii), is encoded by the CD74 gene (DHLAG) and functions as a critical chaperone in MHC class II antigen processing and presentation.
- It binds to peptide-free MHC class II alpha/beta heterodimers, forming a stable alpha-beta-CLIP complex that prevents premature peptide loading, ensuring proper transport from the endoplasmic reticulum to the endosomal/lysosomal system where antigen loading occurs.
- CD74 acts as a chaperone for cathepsin L (CTSL), stabilizing the mature enzyme in endocytic compartments and extracellular spaces of antigen-presenting cells (APCs).
- The p41 isoform exhibits direct antiviral activity by blocking endosomal entry of Ebola virus and coronaviruses, including SARS-CoV-2, through disruption of cathepsin-mediated viral glycoprotein processing (PubMed:32855215).

- The immunogen corresponds to the N-terminal region (amino acids 1–100) of human CD74; consider that this antibody may recognize both isoforms and cleaved forms containing this epitope.
- For Western blot, a band around 34 kDa (consistent with the full-length protein) is expected; additional bands may appear due to proteolytic processing or alternative isoforms.
- In immunohistochemistry (IHC-P), focus on membrane and intracellular vesicular staining patterns in immune tissues, and validate with appropriate controls.
- ELISA performance should be optimized using recombinant CD74 protein or the synthetic peptide immunogen as a coating antigen.
