Biological Background: CD73/NT5E Function and Localization
- CD73/NT5E encodes the ecto-5'-nucleotidase enzyme, a glycosylphosphatidylinositol (GPI)-anchored cell membrane protein that catalyzes the hydrolysis of extracellular nucleotide monophosphates, releasing inorganic phosphate and the corresponding nucleoside.
- The enzyme preferentially hydrolyzes AMP, but also accepts IMP, UMP, GMP, CMP, dAMP, dCMP, dTMP, NAD, and NMN as substrates. Related references: PMID:21933152, PMID:22997138, PMID:23142347
- CD73 shows a preference for ribonucleotide monophosphates over their equivalent deoxyribose forms. Related references: PMID:34403084
- The protein is anchored to the outer leaflet of the plasma membrane via a GPI moiety and contains disulfide bonds, N-linked glycosylation sites, and a zinc-binding site that are essential for its structural integrity and catalytic activity.
- Alternative splicing of the NT5E gene produces multiple isoforms, which may differ in their enzymatic properties or subcellular distribution.
- The UniProt record lists 'Disease variant' as a keyword, suggesting that mutations in NT5E may be associated with human diseases.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 32-264), suggesting the antibody can recognize native cell surface CD73 in intact cells and tissues.
- For Western blotting, expect a band near 63 kDa; however, N-linked glycosylation may cause migration to a higher apparent molecular weight; validate in your specific sample system.
- In immunohistochemistry (IHC-P), consider antigen retrieval conditions and include appropriate positive and negative controls, especially in tissues known to express CD73 such as placenta, kidney, and lymphoid organs.
- Cross-reactivity with mouse and rat CD73 is reported; verify expected bands in those species if using non-human samples.
- For ELISA, the antibody can be employed as a capture or detection reagent; pair with a validated monoclonal antibody or another batch for sandwich ELISA optimization.
