- B-cell differentiation antigen CD72, also known as Lyb-2, is encoded by the CD72 gene in humans.
- Functions as a co-receptor of the B-cell receptor (BCR), playing both positive and negative regulatory roles in B-cell functions.
- Recognizes the Sm/ribonucleoprotein (RNP) self-antigen ligand; coligation of CD72 with the BCR inhibits BCR signaling through the recruitment of PTPN6/SHP-1 to the BCR complex.
- Serves as a ligand for CD5, contributing to the maintenance of regulatory T and B-cell homeostasis.
- Localizes to the membrane as a single-pass type II membrane protein with a signal-anchor and a transmembrane helix.
- Predominantly expressed on pre-B-cells and B-cells, but absent from terminally differentiated plasma cells.
