Biological Background: CD71/Transferrin Receptor Function and Localization
- Transferrin receptor protein 1 (TFRC), also known as T9, p90, or CD71, is a transmembrane receptor essential for cellular iron uptake via receptor-mediated endocytosis of transferrin-bound iron. - Positively regulates T and B lymphocyte proliferation by facilitating iron uptake, highlighting its role in immune cell function. Related references: PMID:26642240
- Acts as a lipid sensor: under low dietary stearate (C18:0), TFRC promotes JNK pathway activation, leading to HUWE1-mediated ubiquitination and degradation of mitofusin MFN2, thereby inhibiting mitochondrial fusion. - For immunofluorescence, permeabilization may be necessary to access intracellular epitopes due to the transmembrane nature of the protein; validate in your specific sample system.
- Given the multiple viral entry roles, blocking or detecting TFRC may be useful in infection assays; however, the neutralizing capacity of this polyclonal antibody should be empirically determined.
- The protein weight is approximately 85 kDa, but glycosylation and splice variants may cause slight migration shifts in WB; consider appropriate positive controls.
