Biological Background: CD71/Transferrin Receptor Function and Localization
- CD71, also known as Transferrin receptor protein 1 (TFR1) and commonly designated TFRC, is the primary mediator of cellular iron uptake, essential for diverse physiological processes.
- It binds iron-loaded transferrin at the cell surface and internalizes the complex via clathrin-mediated endocytosis. Following endosomal acidification, iron is released, and the apotransferrin-receptor complex recycles back to the plasma membrane.
Related references: PMID:26214738, PMID:41772062 - CD71 is crucial for erythropoiesis and nervous system development, highlighting its importance in tissues with high iron demand.
- Beyond iron transport, it positively regulates T and B lymphocyte proliferation via iron uptake, linking iron homeostasis to immune function.
Related references: PMID:26642240 - It acts as a lipid sensor, modulating mitochondrial fusion: under low stearate conditions, it activates the JNK pathway to promote MFN2 degradation, whereas high stearate levels lead to TFRC stearoylation and inhibition of this pathway.
Related references: PMID:26214738 - CD71 is exploited as a host entry receptor by numerous pathogens, including new-world arenaviruses, rabies virus, SARS-CoV, MERS-CoV, and SARS-CoV-2, which hijack its endocytic pathway, and by the malaria parasite Plasmodium vivax.
- The receptor localizes predominantly to the cell membrane and melanosomes, and a secreted form also exists. It is a disulfide-linked homodimeric glycoprotein subject to palmitoylation, phosphorylation, and stearoylation, modifications that regulate its trafficking and functions.
Experimental Guidance and Technical Tips
- This rabbit monoclonal antibody was generated using recombinant human CD71 protein as immunogen; it is expected to recognize endogenous CD71 in human and mouse samples.
- For Western blotting, the expected molecular weight is approximately 85 kDa. Include appropriate molecular weight markers and validate band specificity in your sample system.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and blocking conditions may require optimization for different tissue or cell types.
- As CD71 is a cell surface receptor, live-cell staining protocols may be applicable; confirm compatibility with your experimental design.
- Always include positive and negative controls, and consider testing the antibody in your specific application with a dilution series to determine optimal working concentration.
