Biological Background: CD71/Transferrin Receptor Function and Localization
- Transferrin receptor protein 1 (TfR1)/CD71 is the primary mediator of cellular iron uptake. It binds diferric transferrin and internalizes the complex via clathrin-mediated endocytosis. Endosomal acidification releases iron, and the apotransferrin-receptor complex recycles to the cell surface. TfR1 also binds the hereditary hemochromatosis protein HFE at an overlapping C-terminal site. Related references: PMID:26214738 PMID:41772062
- TfR1 positively regulates T and B cell proliferation through iron uptake. Related references: PMID:26642240
- TfR1 acts as a lipid sensor that modulates mitochondrial fusion. Low dietary stearate levels promote JNK pathway activation and HUWE1-mediated ubiquitination of mitofusin MFN2, inhibiting mitochondrial fusion. High stearate levels lead to TFRC stearoylation, inhibiting JNK activation and stabilizing MFN2. Related references: PMID:26214738
- Serves as a host entry factor for several viruses, including new-world arenaviruses (Guanarito, Junin, Machupo), rabies virus, and coronaviruses (SARS-CoV, MERS-CoV, SARS-CoV-2).
- Acts as a receptor for the malaria parasite Plasmodium vivax.
- Predominantly localized at the cell membrane; also found in melanosomes and secreted.
- Post-translationally modified by palmitoylation and ubiquitination; contains disulfide bonds and is glycosylated. It is a disulfide-linked homodimeric type II transmembrane protein with a signal-anchor domain.
Experimental Guidance and Technical Tips
- For Western blotting, expect a band at approximately 85 kDa, corresponding to the glycosylated TfR1 monomer. Use reducing and denaturing conditions.
- In IHC-P, antigen retrieval (heat-induced at pH 6 or 9) can improve staining. Validate on appropriate positive and negative tissue controls.
- For IF/ICC, cell surface TfR1 can be detected without permeabilization; permeabilization enables staining of intracellular pools. Consider live-cell staining for surface expression.
- This rabbit monoclonal antibody was raised against amino acids 100–250 of human TfR1. Cross-reactivity with other species is not reported, so validate in your sample system if used beyond human.
- Validated for ELISA; consider pairing with a non-competing antibody for sandwich assay development.
