Biological Background: CD69 Function and Localization

  • CD69, also known as AIM, EA1, MLR-3, and CLEC2C, is a type II transmembrane glycoprotein belonging to the C-type lectin domain family.
  • It is rapidly upregulated on the surface of activated platelets, T-lymphocytes, and NK cells in response to antigen recognition or cytokine signaling, contributing to immune homeostasis. Related references: PMID:24752896, PMID:26296369, PMID:35930205
  • CD69 negatively regulates Th17 cell differentiation by interacting with galectin-1 on immature dendritic cells. Related references: PMID:24752896
  • It serves as an oxidized LDL receptor on CD4 T-cells, triggering PDCD1 expression through NFAT to limit inflammatory responses. Related references: PMID:35930205
  • CD69 recognizes Pseudomonas aeruginosa, promoting ERK1 activation and GM-CSF secretion to enhance antibacterial defense. Related references: PMID:34841721
  • Subcellularly, CD69 is anchored in the cell membrane.
  • Its tissue specificity includes expression on activated T-cells, B-cells, NK cells, neutrophils, eosinophils, epidermal Langerhans cells, and platelets.

Experimental Guidance and Technical Tips

  • Given the immunogen is a recombinant fusion protein spanning amino acids 62-199 (the extracellular domain), this antibody is expected to recognize the native conformation of CD69 on the cell surface. Consider using it for flow cytometry applications in addition to the validated applications.
  • For Western blot, a band at approximately 23 kDa (possibly glycosylated forms at higher molecular weights) should be observed under reducing conditions. Validate in your sample system.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), antigen retrieval and detection conditions should be optimized; CD69 is expressed on activated immune cells, so positive controls like activated PBMCs or lymphoid tissue sections are recommended.
  • For ELISA, the antibody can be used as a coating or detection reagent, but appropriate antibody pairing and assay conditions should be determined empirically.
