Biological Background: CD69 Function and Localization

  • CD69 is a type II transmembrane C-type lectin receptor, also known as AIM, EA1, MLR-3, CLEC2C, GP32/28, BL-AC/P26, and Leu-23. It is rapidly upregulated on the surface of activated T-cells, B-cells, natural killer cells, neutrophils, eosinophils, epidermal Langerhans cells, and platelets.
  • It negatively regulates Th17 cell differentiation through carbohydrate-dependent interaction with galectin-1/LGALS1 on immature dendritic cells. Related references: PMID:24752896
  • CD69 cooperates with the S100A8/S100A9 complex present on peripheral blood mononuclear cells to promote the conversion of naive CD4 T-cells into regulatory T-cells. Related references: PMID:26296369
  • As an oxidized low-density lipoprotein (oxLDL) receptor in CD4 T-lymphocytes, it negatively regulates inflammation by inducing PDCD1 expression through NFAT activation. Related references: PMID:35930205
  • In adipose tissue-derived mesenchymal stem cells (ASCs), CD69 specifically recognizes P. aeruginosa and triggers ERK1 activation, leading to granulocyte-macrophage colony-stimulating factor (GM-CSF) and inflammatory cytokine secretion, thus contributing to host defense. Related references: PMID:34841721
  • CD69 negatively regulates the chemotactic responses of effector lymphocytes and dendritic cells to sphingosine 1-phosphate (S1P) by acting as a S1PR1 agonist and facilitating receptor internalization and degradation. Related references: PMID:37039481
  • The protein resides primarily on the cell membrane as a disulfide-linked homodimer and undergoes post-translational modifications, including glycosylation and phosphorylation.
  • It is expressed on various immune cell subsets and plays a role in eosinophil IL-10 production via the ERK1/2 pathway, though further validation in specific model systems is advised.

Experimental Guidance and Technical Tips

  • The immunogen encompasses amino acids 62–199 of human CD69 (NP_001772.1), corresponding to the extracellular domain. This region is conserved between human and rat, explaining the observed cross-reactivity.
  • For western blot (WB), a band around 23 kDa is expected under reducing conditions; however, due to glycosylation, a higher molecular weight may be observed. Include appropriate positive and negative controls in your experimental design.
  • In ELISA applications, consider direct, indirect, or sandwich formats depending on assay requirements. Titrate the antibody to determine the optimal working concentration for your specific protocol.
  • As CD69 is transiently expressed upon stimulation, activation of target cells (e.g., with PMA/ionomycin for T-cells) may be necessary to detect the protein. Validate detection conditions in your relevant biological samples.
