Biological Background: CD69 Function and Localization
- CD69, also designated activation inducer molecule (AIM), early T-cell activation antigen p60, or Leu-23, is a transmembrane C-type lectin domain family 2 member C (CLEC2C) encoded by the CD69 gene in humans.
- Serves as an early activation marker rapidly upregulated on the surface of platelets, T-lymphocytes, and NK cells following antigen recognition or cytokine signaling, playing a central role in immune cell homeostasis.
- Negatively regulates Th17 cell differentiation through carbohydrate-dependent interaction with galectin-1 (LGALS1) present on immature dendritic cells, thereby modulating JAK3/STAT5 signaling and RORγt transcription. Related references: PMID:24752896
- Promotes the conversion of naïve CD4 T-cells into regulatory T-cells (Tregs) by engaging the S100A8/S100A9 complex on peripheral blood mononuclear cells (PBMCs). Related references: PMID:26296369

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–80), and as a polyclonal antibody, it recognizes multiple epitopes. Consider validating specificity using activated vs. resting lymphocyte lysates.
- For Western blotting, a band at approximately 23 kDa (under reducing conditions) is expected. Use appropriate positive controls such as PHA-stimulated PBMCs or Jurkat T-cell lysates.
- In ELISA, the antibody may be suitable for detecting soluble CD69 shed from activated cells or for cell-based assays; titration and optimization are recommended.
- Cross-reactivity with mouse and rat has been reported; however, confirm reactivity patterns in your specific experimental setup.
