  • CD68 (macrosialin, Gp110) is a heavily glycosylated type I transmembrane protein that belongs to the lysosome-associated membrane protein (LAMP) family.
  • It plays a key role in the phagocytic activities of tissue macrophages, participating in both intracellular lysosomal metabolism and extracellular cell–cell or cell–pathogen interactions.
  • CD68 binds to tissue- and organ-specific lectins or selectins, enabling homing of macrophage subsets to particular sites within the body.
  • The protein rapidly recycles between endosomes/lysosomes and the plasma membrane, a dynamic process that may facilitate macrophage crawling over selectin-bearing substrates or other cells.
  • Subcellularly, CD68 localizes primarily to the cell membrane, endosome membrane, and lysosome membrane.
  • It is highly expressed in blood monocytes and tissue macrophages, but can also be found in lymphocytes, fibroblasts, endothelial cells, and many tumor cell lines.

  • For immunohistochemistry on paraffin-embedded tissues (IHC-P), consider antigen retrieval (e.g., heat-mediated citrate buffer, pH 6.0) and optimize the primary antibody dilution empirically.
  • Cross-reactivity with mouse CD68 has been reported, allowing potential use in mouse models; however, validation in the relevant species and sample type is recommended.
