Biological Background: CD68 Function and Localization

  • CD68 is a heavily glycosylated integral membrane protein belonging to the lysosome-associated membrane protein (LAMP) family, also known as macrosialin or Gp110.
  • It plays a role in phagocytic activities of tissue macrophages, both in intracellular lysosomal metabolism and extracellular cell-cell and cell-pathogen interactions.
  • CD68 binds to tissue- and organ-specific lectins or selectins, allowing homing of macrophage subsets to particular tissue sites.
  • Rapid recirculation of CD68 from endosomes and lysosomes to the plasma membrane may allow macrophages to crawl over selectin-bearing substrates or other cells.
  • The protein is localized primarily to endosome and lysosome membranes, with transient expression on the cell membrane.
  • Highly expressed in blood monocytes and tissue macrophages, and also found in lymphocytes, fibroblasts, endothelial cells, and many tumor cell lines.
  • Its N-terminal mucin-like domain is heavily O-glycosylated, and the protein contains a single transmembrane helix and a short cytoplasmic tail.
