  • CD68, also known as macrosialin, GP110, LAMP4, or SCARD1, is a heavily glycosylated transmembrane glycoprotein belonging to the LAMP family.
  • It is predominantly expressed on the membrane of endosomes and lysosomes, but can also rapidly recycle to the cell surface, suggesting a role in adhesion and cellular motility.
  • CD68 functions in the phagocytic activities of macrophages, participating in both intracellular lysosomal metabolism and extracellular cell–cell and cell–pathogen interactions.
  • By binding to tissue- and organ-specific lectins or selectins, CD68 facilitates the homing of macrophage subsets to specific anatomical sites.
  • The protein contains a mucin-like domain that is likely involved in ligand binding and may mediate attachment to selectins on endothelial cells, aiding in the dissemination of macrophages and tumor cells.

  • For Western blot (WB), it is advisable to test both reducing and non-reducing conditions, and note that the observed band may migrate between 37 kDa (core protein) and 110 kDa (glycosylated form), depending on the sample and experimental conditions.
  • For immunofluorescence/immunocytochemistry (IF/ICC), expect a staining pattern consistent with endosomes, lysosomes, and the cell membrane; fixation and permeabilization protocols should be optimized for the cell type.
