- Macrosialin (CD68), also known as LAMP4, gp110, or SCARD1, is encoded by the Cd68 gene in mouse. Alternative splicing produces multiple isoforms.
- It functions in phagocytic activities of tissue macrophages, including intracellular lysosomal metabolism and extracellular cell-cell and cell-pathogen interactions.
- CD68 rapidly recirculates between endosomes/lysosomes and the plasma membrane, which may facilitate macrophage crawling over selectin-bearing substrates.
- The protein is a heavily glycosylated transmembrane glycoprotein with a signal peptide and a disulfide bond, localized mainly to endosome and lysosome membranes, but also detected at the cell membrane.
- Expressed predominantly in tissue macrophages, with lower expression in dendritic cells.
- Through binding to tissue- and organ-specific lectins or selectins, CD68 contributes to macrophage subset homing to particular sites.
