Biological Background: CD66a/CEACAM1 Function and Localization
- CEACAM1 (CD66a) is a member of the carcinoembryonic antigen (CEA) family and functions as a homophilic, calcium-independent cell adhesion molecule.
- Acts as a coinhibitory receptor in T cells, NK cells, and neutrophils; phosphorylation-dependent PTPN6 recruitment inhibits TCR-mediated cytotoxicity, T cell proliferation, and cytokine production. Related references: PMID:18424730 PMID:23696226 PMID:25363763
- Inhibits cell migration and scattering by binding to FLNA, disrupting the FLNA-RALA interaction. Related references: PMID:16291724
- Regulates insulin homeostasis by promoting insulin clearance and modulating hepatic fatty acid synthesis via INSR-mediated phosphorylation and FASN interaction.
- Functions in angiogenesis and arteriogenesis, promoting endothelial cell differentiation, migration, and vascular remodeling post-ischemia.
- Primarily localized to the plasma membrane in multiple domains (lateral, apical, basal) and cell junctions (adherens junctions); also found in cytoplasmic vesicles, microvilli, and secreted forms.
- Expressed in colon epithelial cells, granulocytes, lymphocytes, and T cells, with alternative splicing generating isoforms with differential cytoplasmic domain lengths.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering human and mouse CEACAM1; cross-reactivity with both species is reported. For species-specific applications, confirm reactivity in the appropriate lysate or tissue.
- Due to alternative splicing and glycosylation, CEACAM1 exhibits multiple isoforms with molecular weights from ~30 kDa to 57 kDa. In Western blot, expect multiple bands; validate against known positive controls.
- For immunohistochemistry (IHC), colon epithelium is a suitable positive control tissue as CEACAM1 is highly expressed in columnar epithelial cells.
- In immunofluorescence (IF), the protein localizes to cell membranes and junctions; use appropriate fixation and permeabilization protocols to detect surface and intracellular pools.
- For ELISA, consider pairing with a capture antibody targeting a different epitope to avoid competition from soluble CEACAM1 forms.
