Biological Background: CD62P/P-selectin Function and Localization
- CD62P/P-selectin (encoded by the SELP gene) is a Ca²⁺-dependent adhesion receptor belonging to the selectin family, which also includes E-selectin and L-selectin.
- It mediates the initial tethering and rolling of leukocytes on activated endothelium or platelets by binding to sialyl-Lewis X carbohydrates on glycoproteins such as PSGL-1 (SELPLG).
- P-selectin also contributes to stable cell adhesion and signaling through interactions with integrins alpha-IIb/beta3 (ITGA2B:ITGB3) and alpha-V/beta-3 (ITGAV:ITGB3). Related references: PMID:37184585
- The protein is primarily stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells. Upon cell activation by agonists like thrombin or histamine, it is rapidly translocated to the plasma membrane.

- For Western blot (WB), the predicted molecular weight is ~91 kDa, though post-translational modifications such as glycosylation may lead to a higher apparent mass. Use reducing conditions and validate in cell lysates known to express P-selectin, such as activated platelets or endothelial cells.
- In immunohistochemistry (IHC-P), P-selectin expression is typically low in resting tissues. Consider using antigen retrieval methods and including positive controls like inflamed or thrombotic tissue sections.
