Biological Background: P-selectin Function and Localization

  • P-selectin (SELP), also designated CD62P, GMP-140, LECAM3, and PADGEM, is a member of the selectin family of cell adhesion molecules. It is a type I transmembrane glycoprotein featuring an N-terminal C-type lectin domain, an EGF-like domain, multiple sushi (SCR) repeats, and a cytoplasmic tail.
  • Functions as a Ca²⁺-dependent receptor that binds carbohydrate ligands, primarily sialyl-Lewis X, on myeloid cells such as neutrophils and monocytes. This interaction mediates the initial tethering and rolling of leukocytes on activated endothelium and platelets during inflammation.
  • Through its counter-receptor SELPLG (PSGL-1) on leukocytes, P-selectin supports leukocyte rolling along vascular surfaces, a critical prerequisite for firm adhesion and extravasation.
  • Additionally, P-selectin facilitates cell–cell interactions and adhesion by engaging integrin αIIb/β3 (ITGA2B:ITGB3) and αV/β3 (ITGAV:ITGB3), contributing to platelet–leukocyte aggregate formation. Related references: PMID:37184585
  • Subcellularly, P-selectin is stored in the α-granules of platelets and Weibel–Palade bodies of endothelial cells. Upon cellular activation by agonists such as thrombin or histamine, it is rapidly translocated to the plasma membrane.
  • Post‑translational modifications include disulfide bond formation, palmitoylation on cytoplasmic cysteine residues, and N‑linked glycosylation. Keywords: Cell adhesion, Calcium, Disulfide bond, Glycoprotein, Lipoprotein, Membrane, Metal-binding.
  • The protein has a calculated molecular weight of approximately 91 kDa and may exhibit mobility shifts on SDS-PAGE due to heterogeneous glycosylation.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 42–771 of human P-selectin, corresponding to nearly the entire extracellular region. This design favors recognition of native conformations in cell‑based applications.
  • For Western blotting, consider using reducing conditions and a molecular weight standard near 91 kDa. Due to glycosylation, the band may appear diffuse or migrate slightly higher.
  • In immunohistochemistry (paraffin), antigen retrieval (heat‑ or enzyme‑based) is recommended; validate antibody specificity on inflamed tissue where P-selectin expression is upregulated.
  • For immunofluorescence, permeabilization enables detection of both intracellular stores and surface‑translocated protein. Co‑localization with organelle markers (e.g., von Willebrand factor for Weibel–Palade bodies) can be instructive.
  • As a rabbit monoclonal, this antibody is compatible with multiplex staining protocols using mouse primary antibodies.
