Biological Background: P-selectin Function and Localization
- P-selectin (CD62P, GMP-140, LECAM3, PADGEM) is a transmembrane adhesion molecule of the selectin family, encoded by the SELP gene.
- It functions as a Ca²⁺-dependent receptor that mediates the initial tethering and rolling of leukocytes on activated endothelium or platelets by binding to sialyl-Lewis X carbohydrates on neutrophils and monocytes.
- Through its interaction with integrins alpha-IIb/beta3 (ITGA2B:ITGB3) and alpha-V/beta-3 (ITGAV:ITGB3), P-selectin facilitates cell-cell adhesion and stable leukocyte recruitment. Related references: PMID:37184585
- P-selectin is stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells and is rapidly translocated to the cell surface upon cellular activation by agonists.
- Tissue specificity is primarily in platelets and endothelial cells, where it is mobilized to the surface during inflammatory responses.
- The protein contains EGF-like and sushi domains, disulfide bonds, and palmitoylation sites, and it is classified as a lectin and calcium-binding membrane glycoprotein.

Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning amino acids 42–771, covering most of the extracellular region; this may allow recognition of native and recombinant P-selectin in various assay formats.
- For Western blotting, consider testing both reducing and non-reducing conditions, as P-selectin is glycosylated and may exhibit an altered apparent molecular weight.
- In immunofluorescence/immunocytochemistry (IF/ICC), appropriate permeabilization and fixation conditions should be optimized, given that P-selectin is surface-exposed upon cellular activation.
- For ELISA applications, titration of the antibody and validation with relevant standards or controls is recommended to ensure sensitivity and specificity.
