Biological Background: CD62P/P-selectin Function and Localization
- CD62P, also known as P-selectin, GMP-140, LECAM3, or PADGEM, is encoded by the SELP gene and belongs to the selectin family of cell adhesion molecules.
- Functions as a Ca2+-dependent receptor that binds sialyl-Lewis X carbohydrates on neutrophils and monocytes, mediating leukocyte rolling on activated endothelial cells or platelets, and also mediates cell-cell interactions and adhesion via integrins alpha-IIb/beta3 and alpha-V/beta3 (PubMed:37184585). - In resting cells, P-selectin is stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells; upon cell activation, it is rapidly transported to the cell surface to mediate adhesive interactions.
- The protein contains structural domains including a C-type lectin domain, an EGF-like domain, and multiple Sushi (CCP/SCR) repeats, along with a transmembrane helix and a short cytoplasmic tail; it undergoes glycosylation, palmitoylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 220–319 of human CD62P (NP_002996.2), a region within the extracellular domain. For Western blotting, a band around 91 kDa is expected; use reducing conditions and appropriate whole-cell lysates from human platelets or endothelial cells (HUVEC) as positive controls.
- For ELISA, consider using the purified recombinant protein or cell-based assays with activated platelets/endothelial cells; validate antibody specificity with appropriate negative controls.
- Cross-reactivity with mouse P-selectin has been observed; verify performance in the relevant mouse tissue or cell system before large-scale experiments.
