Biological Background: CD62P/P-selectin Function and Localization
- P-selectin (CD62P), also known as GMP-140, PADGEM, and LECAM3, is a transmembrane glycoprotein encoded by the SELP gene. It belongs to the selectin family of adhesion molecules (UniProt: P16109).
- It functions as a Ca²⁺-dependent receptor that mediates the initial rolling adhesion of leukocytes on activated endothelium or platelets by binding sialyl-Lewis X carbohydrates on neutrophils and monocytes.
- P-selectin mediates cell-cell interactions and cell adhesion via interaction with integrin alpha-IIb/beta3 (ITGA2B:ITGB3) and integrin alpha-V/beta-3 (ITGAV:ITGB3). Related references: PMID:37184585
- The protein is stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells, and is rapidly translocated to the cell surface upon activation by agonists such as thrombin or histamine.
- Structurally, P-selectin contains an N-terminal lectin domain, an EGF-like domain, multiple Sushi (SCR) repeats, a transmembrane helix, and a short cytoplasmic tail. It is modified by palmitoylation and disulfide bond formation.
- Key biological roles highlighted by UniProt keywords: cell adhesion, calcium binding, lectin activity, and involvement in the inflammatory response.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 731-830 of human P-selectin (P16109), suggesting the antibody likely targets an epitope in the C-terminal region. Consider this when designing complementary detection tools.
- For Western blot, the predicted molecular weight is approximately 91 kDa. Validate using positive controls such as activated platelet or endothelial cell lysates, and optimize antibody dilution empirically.
- In IHC-P, antigen retrieval may be required due to the transmembrane nature of the target. Use appropriate tissue sections known to express P-selectin (e.g., inflamed vascular endothelium).
- For ELISA applications, consider using recombinant P-selectin protein or cell-based assays. Pairing with a detection antibody targeting a different epitope may improve assay performance.
