Biological Background: CD62E Function and Localization
- E-selectin (CD62E/ELAM-1/LECAM2) is a cell-surface glycoprotein encoded by the Sele gene in mice.
- It mediates the adhesion of blood neutrophils to cytokine-activated endothelium by interacting with SELPLG/PSGL1. Related references: PMID:1375914
- Belongs to the selectin family, characterized by an N-terminal lectin domain, an EGF-like domain, and multiple Sushi (SCR) repeats.
- Localized to the cell membrane as a single-pass type I membrane protein.
- Contains structural features including disulfide bonds, a calcium-dependent lectin domain, and N-glycosylation sites.
- May also contribute to capillary morphogenesis.
- Molecular weight is approximately 67 kDa (calculated) or 66 kDa (observed).
Experimental Guidance and Technical Tips
- The immunogen covers the entire extracellular region (residues 22–557), which contains the lectin, EGF-like, and Sushi domains; it may be effective for recognizing both membrane-bound and shed soluble forms.
- For western blot, a specific band at the expected molecular weight (~66 kDa) should be observed; optimization of blocking and antibody dilution is recommended.
- For immunofluorescence and immunocytochemistry, fixation and permeabilization conditions should be optimized, as the target is a transmembrane protein.
- For flow cytometry, using a live-cell staining protocol is suggested to detect surface expression; include appropriate isotype controls.
