Biological Background: CD55 Function and Localization

  • CD55, also known as complement decay-accelerating factor (DAF), is a glycoprotein that regulates the complement cascade by binding C4b and C3b fragments and accelerating the decay of C3 and C5 convertases, thereby protecting host cells from autologous complement attack.
    Related references: PMID:7525274 PMID:28657829
  • The protein acts as a receptor for several viruses, including Coxsackievirus A21, coxsackieviruses B1, B3, and B5, as well as human echoviruses 6, 7, 11, 12, 20, and 21, and enteroviruses 70 and D68, facilitating viral entry into host cells.
  • CD55 is broadly expressed on plasma membranes of all cell types exposed to plasma complement proteins and on epithelial cells lining extracellular compartments, with soluble forms present in body fluids and extracellular matrix.
  • Encoded by the CD55 gene (Gene ID: 1604), the protein undergoes alternative splicing, generating multiple isoforms, some of which are secreted.
  • CD55 is attached to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor and contains multiple sushi domains; it is also a carrier of the Cromer blood group system antigens.
  • Post-translational modifications include extensive N-glycosylation and formation of disulfide bonds, which are critical for structural stability and function.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 40–300 of human CD55, corresponding to the extracellular domain; for Western blot, a band near 41 kDa is expected under reducing conditions, though glycosylation may cause subtle shifts.
  • This polyclonal antibody is listed for WB, IF/ICC, and ELISA; optimal dilutions should be empirically determined for each assay and sample type.
  • Cross-reactivity with mouse and rat is specified; sequence homology in the immunogen region should be verified if quantitation across species is required.
  • For IF/ICC, consider permeabilization conditions as CD55 is a GPI-anchored surface protein; include appropriate controls to confirm membrane localization.
