Biological Background: CD50 (ICAM-3) Function and Localization

• CD50, also known as ICAM-3 or CDw50, is a member of the intercellular adhesion molecule (ICAM) family and functions as a ligand for the integrin LFA-1 (ITGAL/ITGB2), mediating leukocyte adhesion and facilitating apoptotic neutrophil phagocytosis by macrophages. • Titrate the antibody to determine the optimal staining concentration for your specific cell type and experimental conditions. A starting range of 0.5–5 µg per million cells is typical for conjugate antibodies.
• Since CD50 is expressed on all resting leukocytes, validate the antibody on known positive and negative cell populations (e.g., peripheral blood lymphocytes vs. endothelial cells).
• For intracellular staining, a fixation/permeabilization step is required; however, note that this antibody targets an extracellular epitope and may perform best on surface staining of unfixed cells.
