Biological Background: CD50 (ICAM-3) Function and Localization

  • CD50, also known as ICAM-3 or CDw50, is a member of the intercellular adhesion molecule (ICAM) family and functions as a ligand for the integrin LFA-1 (ITGAL/ITGB2), mediating leukocyte adhesion and facilitating apoptotic neutrophil phagocytosis by macrophages.   • It serves as a ligand for integrin ITGAD/ITGB2, contributing to cell-cell interactions within the immune system. Related references: PMID:8777714
  • CD50 binds to CD209 on dendritic cells through a Ca²⁺-dependent, integrin-independent mechanism, establishing initial contact between dendritic cells and resting T cells and promoting T cell proliferation. Related references: PMID:10721994

Experimental Guidance and Technical Tips

  • The immunogen was designed against the extracellular domain (amino acids 30–114), so this antibody is expected to recognize the native surface conformation of CD50 on live cells. Consider using appropriate isotype controls and viability dyes in flow cytometry to gate on live, single cells.
  • Titrate the antibody to determine the optimal staining concentration for your specific cell type and experimental conditions. A starting range of 0.5–5 µg per million cells is typical for conjugate antibodies.
  • Since CD50 is expressed on all resting leukocytes, validate the antibody on known positive and negative cell populations (e.g., peripheral blood lymphocytes vs. endothelial cells).
  • For intracellular staining, a fixation/permeabilization step is required; however, note that this antibody targets an extracellular epitope and may perform best on surface staining of unfixed cells.
