Biological Background: CD5 Function and Localization
- CD5, also referred to as T1 or Leu-1, is a lymphoid-specific transmembrane receptor characterized by three extracellular scavenger receptor cysteine-rich (SRCR) domains, a single transmembrane helix, and a cytoplasmic tail involved in signal transduction.
- It is predominantly expressed on all T-cells and a subset of B1a B-cells, playing a crucial role in modulating T-cell receptor (TCR) and B-cell receptor (BCR) signaling, thymocyte selection, T-cell effector differentiation, and immune tolerance.
- CD5 serves as a negative regulator of TCR signaling by interacting with key signaling molecules such as LCK, CD3ζ, PI3K, and CBL. Upon co-engagement of CD3, phosphorylation of CBL is enhanced, which promotes VAV1 phosphorylation and subsequent degradation. Related references: PMID:1384049 PMID:1385158 PMID:23376399
- Through interactions with ligands like CD5L and CD72 on B-cells, CD5 facilitates contact-mediated, T-dependent B-cell activation and helps maintain regulatory T and B-cell homeostasis.
- In B-cells, CD5 signaling leads to ERK1/2 activation via the calcium channel TRPC1, resulting in IL-10 production and modulation of B-cell effector functions. Related references: PMID:27499044
- The CD5 protein localizes to the cell membrane and undergoes post-translational modifications including N-linked glycosylation and phosphorylation, which regulate its stability and signaling activity.
- Keywords associated with CD5 include receptor, immune response, disulfide bond, glycoprotein, and phosphoprotein, underscoring its multifaceted role in immune regulation.
Experimental Guidance and Technical Tips
- The immunogen used to generate this rabbit polyclonal antibody corresponds to amino acids 158–371 of human CD5, which spans a portion of the extracellular domain. This design may favor detection of surface CD5 in immunohistochemistry (IHC-P) applications.
- For Western blot (WB), a band at approximately 55 kDa is expected. Optimization of sample preparation, loading amount, and antibody dilution (e.g., 1:500–1:2000) is recommended using positive control cell lines such as Jurkat (human T-cell leukemia) or mouse splenocytes.
- In IHC-P, antigen retrieval method and antibody titration should be empirically determined. Cross-reactivity with mouse and rat CD5 allows the antibody to be used in common rodent model systems.
- For ELISA, the antibody may be used as a capture or detection reagent. Consider pairing with a matched antibody pair if quantitative analysis is required. Validate matrix effects and specificity using recombinant CD5 protein or cell lysates.
