Biological Background: CD5 Function and Localization

  • CD5 (T-cell surface glycoprotein CD5, also known as T1/Leu-1) is a lymphoid-specific receptor belonging to the scavenger receptor cysteine-rich (SRCR) superfamily. It is expressed on all T cells and on a subset of B cells known as B1a cells.
  • CD5 acts as a negative regulator of T-cell receptor (TCR) signaling during thymocyte development by associating with signaling molecules such as LCK, CD3Z, PI3K, and CBL. This modulation is crucial for appropriate thymocyte selection and T-cell effector differentiation. Related references: PMID:1384049 PMID:1385158
  • Co-engagement of CD3 with CD5 enhances the recruitment of phosphorylated CBL, leading to increased VAV1 phosphorylation and subsequent degradation, thereby fine-tuning TCR signal strength. Related references: PMID:23376399
  • CD5 maintains regulatory T and B-cell homeostasis and participates in contact-mediated T-dependent B-cell activation by interacting with B-cell ligands such as CD5L and CD72.
  • In B cells, CD5 modulates cellular responses through a Ca2+-dependent pathway involving TRPC1, leading to ERK1/2 activation and IL-10 production. Related references: PMID:27499044
  • The CD5 protein is a single-pass type I transmembrane glycoprotein localized at the cell membrane. It contains multiple SRCR domains in its extracellular region and a cytoplasmic tail that mediates its signaling functions.
  • Keywords associated with CD5 include 3D-structure, disulfide bond, glycoprotein, membrane, phosphoprotein, receptor, repeat, signal, and transmembrane helix, reflecting its structural complexity and role in cell signaling.

Experimental Guidance and Technical Tips

  • This antibody is raised against a recombinant fusion protein corresponding to the extracellular region of human CD5 (amino acids 25–371). It is designed for direct flow cytometry detection via the fluorescent label.
  • For flow cytometry, use standard surface staining protocols on live human peripheral blood mononuclear cells (PBMCs) or lymphoid cell lines expressing CD5. Consider titrating the antibody to determine the optimal concentration for your specific cell population.
  • Include appropriate compensation controls when analyzing multiplex panels, as emits in the FITC channel. Isotype controls or fluorescence-minus-one (FMO) controls are recommended for accurate gating.
  • Cell fixation and permeabilization may alter the epitope accessibility; validation under your experimental conditions is advised.
