Biological Background: CD5 Function and Localization

  • T-cell surface glycoprotein CD5 (also known as Leu-1 or T1) is a member of the scavenger receptor cysteine-rich (SRCR) superfamily and functions as a lymphocyte-specific co-receptor.
  • CD5 negatively regulates TCR signaling in thymocytes by recruiting signaling proteins such as LCK, CD3Z chain, PI3K, and CBL. Related references: PMID:1384049, PMID:1385158
  • Co-ligation of CD3 and CD5 enhances CBL phosphorylation, leading to increased VAV1 phosphorylation and degradation, thereby modulating signal strength. Related references: PMID:23376399
  • In B1a cells, CD5 promotes ERK1/2 activation through TRPC1-mediated Ca2+ influx, driving IL-10 production and contributing to regulatory B-cell function. Related references: PMID:27499044
