Biological Background: CD5 Function and Localization
- T-cell surface glycoprotein CD5 (also known as Leu-1 or T1) is a member of the scavenger receptor cysteine-rich (SRCR) superfamily and functions as a lymphocyte-specific co-receptor.
- CD5 negatively regulates TCR signaling in thymocytes by recruiting signaling proteins such as LCK, CD3Z chain, PI3K, and CBL. Related references: PMID:1384049, PMID:1385158
- Co-ligation of CD3 and CD5 enhances CBL phosphorylation, leading to increased VAV1 phosphorylation and degradation, thereby modulating signal strength. Related references: PMID:23376399
- In B1a cells, CD5 promotes ERK1/2 activation through TRPC1-mediated Ca2+ influx, driving IL-10 production and contributing to regulatory B-cell function. Related references: PMID:27499044
- CD5 is expressed on the plasma membrane of all mature T cells and a subset of B cells, with essential roles in immune tolerance and T-dependent B-cell activation.
- It is a glycoprotein with intrachain disulfide bonds, a single-pass transmembrane domain, and a cytoplasmic tail subject to phosphorylation.
- The predicted molecular mass is approximately 54.6 kDa, consistent with the 54 kDa band observed by western blotting.
Experimental Guidance and Technical Tips
- The conjugate is designed for direct flow cytometry; consider titrating from 0.1–1 µg per test using human PBMCs or purified T cells.
- Use appropriate isotype controls and fluorescence compensation settings to ensure signal specificity.
- Validate the antibody in your sample matrix and fixation/permeabilization protocol, as antigen accessibility may vary.
- Cross-reactivity is expected with human CD5 only; confirm if other species are required.
