Biological Background: CD48 Function and Localization

  • CD48, also known as SLAMF2 or BLAST-1, is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein belonging to the signaling lymphocytic activation molecule (SLAM) family.
  • It modulates immune responses by engaging the receptors CD2 and 2B4/CD244, which are critical for T-cell and natural killer (NK) cell activation (PubMed:9881969, PubMed:21278219). Related references: PMID:9881969, PMID:21278219
  • Upon CD2 ligation, CD48 recruits the Src kinase LCK and the linker LAT to the TCR/CD3 complex, thereby enhancing tyrosine phosphorylation and downstream signaling cascades (PubMed:9881969). Related references: PMID:9881969
  • Interaction with 2B4/CD244 triggers phosphorylation of immunoreceptor tyrosine-based switch motifs (ITSMs), leading to immunological synapse assembly and polarized exocytosis of cytotoxic granules by effector lymphocytes (PubMed:21278219). Related references: PMID:21278219
  • CD48 is predominantly located in lipid rafts of the plasma membrane and can also be released in a soluble form; its GPI anchor facilitates compartmentalization for efficient signal transduction.
  • Mature CD48 has a molecular weight of ~27 kDa (UniProt: 27383 Da) and contains immunoglobulin-like domains, disulfide bonds, and N-linked glycosylation sites.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 23–216) encompasses the extracellular domain of mouse CD48, which may be suitable for detecting native surface-exposed epitopes in flow cytometry.
  • For flow cytometry, consider titrating the antibody to determine the optimal working concentration and include appropriate isotype controls to assess non-specific binding.
  • In ELISA applications, pairing with a capture antibody or using recombinant CD48 protein as a standard could help develop quantitative detection systems.
  • Take care to preserve membrane integrity during sample preparation if targeting membrane-bound CD48, as its GPI anchor may be sensitive to certain detergents.
