Biological Background: CD47 Function and Localization
- CD47, also known as integrin-associated protein (IAP), is a ubiquitously expressed transmembrane glycoprotein of the immunoglobulin superfamily.
- It functions as a receptor for thrombospondin-1 (THBS1), modulating integrin signaling and heterotrimeric G protein activation, and plays a key role in cardiovascular homeostasis, nitric oxide signaling, and blood pressure regulation. Related references: PMID:20610415 PMID:27742621
- Through SIRPA binding, it transmits an anti-phagocytic signal that protects red blood cells from elimination, and by similarity, suppresses dendritic cell activation. Related references: PMID:10856220
- Positively regulates FAS-dependent apoptosis in T-cells by facilitating FAS clustering. Related references: PMID:15917238
- Inhibits stem cell self-renewal and negatively modulates wound healing downstream of THBS1, involving regulation of pluripotency transcription factors POU5F1/OCT4, SOX2, MYC, and KLF4. Related references: PMID:23591719 PMID:18156939
- Suppresses angiogenesis and has been implicated in metabolic dysregulation during normal aging. Related references: PMID:32679764
- Primarily localized to the cell membrane, with an extracellular immunoglobulin V-type domain, multiple transmembrane helices, and a short cytoplasmic tail.
- Post-translational modifications include N-linked glycosylation, phosphorylation, an N-terminal pyrrolidone carboxylic acid, and disulfide bonds, which modulate its structure and ligand interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular region (aa 19–135), which contains the ligand-binding immunoglobulin domain. Epitope preservation in native conformation is critical; users may evaluate the antibody in non-reducing SDS-PAGE for disulfide bond-dependent epitopes.
- For Western blotting, a band around 33 kDa is expected; note that glycosylation may cause the protein to migrate at a slightly higher molecular weight.
- The antibody is validated for detection of mouse and rat CD47; cross-reactivity with other species should be independently verified.
- For ELISA, consider pairing with a detection antibody or using the antibody for direct/indirect detection, and optimize coating conditions for recombinant or native protein.
