Biological Background: CD46 Function and Localization
- CD46, also known as membrane cofactor protein (MCP) or trophoblast leukocyte common antigen (TLX), is a type I membrane glycoprotein belonging to the regulators of complement activation (RCA) gene cluster.
- It serves as a cofactor for the serine protease factor I, enabling the cleavage and inactivation of C3b and C4b deposited on host cells, thereby protecting autologous cells from complement-mediated damage.
- CD46 plays a role in fertilization by participating in sperm-egg fusion and is localized to the inner acrosomal membrane of spermatozoa.
- It acts as a costimulatory molecule for T cells, promoting the differentiation of CD4+ T cells into interleukin-10-secreting T regulatory type 1 (Tr1) cells, which suppress immune responses and help prevent autoimmunity.
- CD46 mediates immune evasion by serving as a receptor for several pathogens, including adenoviruses (subgroup B2/Ad3), measles virus, HHV-6, and pathogenic bacteria such as Neisseria meningitidis and Streptococcus pyogenes. - Expressed on the surface of all nucleated cells except erythrocytes, CD46 is also found in cytoplasmic vesicles and secretory vesicles.
- Genetic variants in CD46 are associated with atypical hemolytic uremic syndrome (AHUS2), underlining its critical role in complement regulation.
- The mature protein has a predicted molecular weight of ~44 kDa, appearing as a 37-44 kDa band in western blotting due to glycosylation.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal extracellular region (aa 35-328), which encompasses multiple sushi domains critical for ligand binding; this may contribute to recognition of native CD46 in applications like WB, IF, and FC.
- For western blot, a band at 37-44 kDa is expected; consider using appropriate positive and negative controls and optimizing lysis buffers and blocking conditions for your sample type.
- In immunofluorescence and flow cytometry, validate the antibody under your specific fixation/permeabilization protocols; note that CD46 is broadly expressed, so adjust cell numbers and staining concentrations accordingly.
- For ELISA applications, verify compatibility with the antigen format (e.g., recombinant protein or cell lysates) and include relevant standard curves.
- As CD46 is also involved in T-cell costimulation, functional assays may require careful titration; consult the literature for assay-specific conditions.
