Biological Background: CD45 Function and Localization
- CD45, also known as Leukocyte common antigen (L-CA) or T200, is a receptor-type tyrosine-protein phosphatase C encoded by the PTPRC gene.
- Functions as a protein tyrosine phosphatase essential for T-cell activation through the antigen receptor, acting as a positive regulator of T-cell coactivation upon binding to DPP4. Related references: PMID:35767951
- Contains two PTPase domains; the first has enzymatic activity, while the second likely modulates substrate specificity.
- Upon T-cell activation, recruits and dephosphorylates SKAP1 and FYN, and modulates LYN activity through dephosphorylation.
- Interacts with CLEC10A at the antigen presenting cell–T cell contact, regulating T cell activation threshold to limit autoreactivity.
- Acts as a receptor for human cytomegalovirus UL11, mediating binding to T-cells and impairing T-cell receptor signaling.
- Localizes to the cell membrane, membrane rafts, and the immunological synapse.
- Multiple isoforms exist through alternative splicing, with tissue-specific expression patterns primarily in thymocytes.
