Biological Background: CD45 Function and Localization

  • CD45 (receptor-type tyrosine-protein phosphatase C), also known as leukocyte common antigen (L-CA), LY5, B220, and T200, is encoded by the PTPRC gene. Multiple isoforms arise from alternative splicing.
  • It is a critical positive regulator of T-cell activation through the antigen receptor. The first phosphatase domain has catalytic activity, while the second modulates substrate specificity. Related references: PMID:35767951
  • CD45 dephosphorylates key signaling molecules: upon T-cell activation, it recruits and dephosphorylates SKAP1 and FYN, and modulates LYN activity. Interaction with CLEC10A at the dendritic cell–T cell interface modulates the activation threshold to limit autoreactivity.
  • It acts as a receptor for human cytomegalovirus protein UL11, mediating binding to T-cells and impairing TCR signaling and proliferation.
  • Subcellularly, CD45 localizes to the cell membrane, membrane rafts, and immunological synapse. It is extensively glycosylated and phosphorylated.
  • Tissue-specific isoform expression: isoforms 1, 2, 3, 5, and 7 are detected in thymocytes, while isoforms 4, 6, and 8 are not.

Experimental Guidance and Technical Tips

  • For Western blot (WB), a band at ~147 kDa is expected; validate the antibody in appropriate leukocyte lysates from human, mouse, or rat.
  • In immunoprecipitation (IP), CD45 enrichment can be achieved; co-immunoprecipitation experiments may require optimization of lysis and washing conditions.
  • For immunofluorescence (IF/ICC and IF-P), fixation and permeabilization protocols should be optimized; CD45 surface staining can be performed without permeabilization.
  • The immunogen (aa 611–730) lies within the extracellular region, suggesting potential suitability for live-cell surface staining, though validation is recommended.
  • Reported cross-reactivity with mouse and rat makes this antibody a candidate for multi-species studies; always verify specificity in the relevant sample system.
