Biological Background: CD45 Function and Localization

  • CD45, also known as Leukocyte common antigen (L-CA), Ly-5, or T200, is a receptor-type tyrosine-protein phosphatase C encoded by the Ptprc gene in mice. It is a transmembrane glycoprotein with a molecular weight of approximately 145 kDa.
  • CD45 is essential for T-cell activation through the antigen receptor. It functions as a positive regulator of T-cell coactivation upon binding to DPP4.
  • The intracellular region contains two tandem protein tyrosine phosphatase (PTP) domains. The first domain possesses catalytic activity, while the second domain modulates substrate specificity.
  • Upon T-cell activation, CD45 recruits and dephosphorylates signaling molecules such as SKAP1 and FYN, and also dephosphorylates LYN to modulate its activity.
  • CD45 interacts with CLEC10A at the contact site between antigen-presenting cells and T cells. This interaction, involving Tn antigen-carrying CD45 on effector T cells and CLEC10A on immature dendritic cells, helps tune T-cell activation thresholds to prevent autoreactivity.
  • The protein localizes to the cell membrane, membrane rafts, and immunological synapses, consistent with its role in proximal signaling. Post-translational modifications include alternative splicing and phosphorylation.

  • The immunogen corresponds to amino acids 26–566 of mouse CD45 (the extracellular region), suggesting the antibody likely recognizes an extracellular epitope. This makes it well-suited for live-cell staining and flow cytometry without permeabilization.
  • For immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), consider optimizing fixation conditions (e.g., 4% PFA) and, if needed, use mild permeabilization to ensure antibody access while preserving epitope integrity.
  • When performing flow cytometry (FC), titrate the antibody to determine the optimal concentration for your specific mouse cell type, and include appropriate isotype controls.
  • This antibody is validated for use with mouse samples. Cross-reactivity with other species has not been reported; verify in your system if needed.
