Biological Background: CD44 Function and Localization
- CD44 (Ly-24, Pgp-1, HERMES) is a cell-surface receptor that mediates cell-cell interactions, adhesion, and migration, and participates in T-lymphocyte activation, recirculation, homing, hematopoiesis, inflammation, and response to bacterial infection. Related references: PMID:25065622 PMID:8343954
- Through its ectodomain, CD44 binds hyaluronan (HA), collagen, growth factors, cytokines, or proteases, and transduces signals by assembling cytoplasmic complexes with PKN2, RhoGTPases (RAC1, RHOA), Rho-kinases, and phospholipase C to regulate calcium mobilization and actin cytoskeleton reorganization.
- Engagement with LGALS9 ligand activates LCK, ERK, and MAPK pathways to promote NK cell activation. Related references: PMID:37006235
- CD44 is expressed in the intestinal epithelium.
- Subcellularly, it localizes to the cell membrane, microvilli, and can be secreted.
- The protein undergoes alternative splicing, phosphorylation, sulfation, and extensive glycosylation, contributing to a broad observed molecular weight range (40-86 kDa).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 23-176) of mouse CD44, targeting a region conserved across human, mouse, and rat, which supports the observed cross-reactivity.
- For Western blotting, note the broad 40–86 kDa range due to heavy glycosylation and splice variants; consider appropriate gel percentages and transfer conditions.
- For IHC-P, antigen retrieval optimization may be necessary; validate with known positive and negative tissue controls.
- Confirm antibody specificity in your experimental system using knockout cell lines or siRNA silencing where feasible.
- This antibody is suitable for WB, IF/ICC, IHC-P, and ELISA; individual optimization of dilutions and conditions is recommended.
