  • CD44 is a highly versatile transmembrane glycoprotein known by alternate names including Ly-24, Pgp-1, HERMES, ECMR-III, and hyaluronate receptor.
  • It functions as a cell-surface receptor mediating cell adhesion, migration, and T-lymphocyte homing by binding hyaluronan, collagen, and other extracellular matrix components.   • CD44 localizes to the plasma membrane and microvilli and is also found as a secreted form; in mouse, it is notably expressed in the intestinal epithelium.
  • The receptor undergoes extensive post-translational modification including N- and O-glycosylation, sulfation, phosphorylation, and proteoglycan substitution, and contains structural disulfide bonds.

  • The immunogen corresponds to the extracellular domain (amino acids 23-176) of mouse CD44. Antibodies generated against this region are expected to recognize the native, surface-exposed receptor.
  • For Western blotting, a band around 86 kDa is predicted; however, extensive glycosylation may cause migration at a higher apparent molecular weight—include proper positive controls and consider deglycosylation if necessary.
  • In immunofluorescence/immunocytochemistry, the antibody should detect membrane-localized CD44; optimize fixation and permeabilization steps to preserve epitope accessibility.
  • For flow cytometry, use standard surface staining protocols and titrate the antibody on the relevant mouse cell population.
  • The antibody is validated for mouse samples; cross-reactivity with other species has not been determined and should be tested if needed.
