Biological Background: CD44 Function and Localization

  • CD44 is a cell-surface receptor, also known as PGP-1, ECMR-III, HUTCH-1, and Epican, encoded by the CD44 gene in humans.
  • It mediates cell-cell and cell-extracellular matrix adhesion by binding hyaluronan and other ligands, playing a crucial role in lymphocyte homing, hematopoiesis, and inflammation. Related references: PMID:16541107 PMID:19703720 PMID:22726066 PMID:7528188
  • Through its cytoplasmic domain, CD44 assembles signaling complexes involving PKN2, RAC1, RHOA, and phospholipase C to regulate calcium mobilization and actin cytoskeletal reorganization. Related references: PMID:15123640
  • Alternative splicing generates multiple isoforms; isoform 10 (epithelial isoform) is highly expressed in carcinomas and repressed in neuroblastoma.
  • Subcellularly, CD44 localizes to the cell membrane, microvilli, and is also found in a secreted form.
  • Post-translational modifications include glycosylation, phosphorylation, and proteoglycan addition; annotated as a glycoprotein, phosphoprotein, and proteoglycan.

Experimental Guidance and Technical Tips

  • For Western blot, a band around 82kDa is expected, but alternative splicing and extensive glycosylation may produce multiple bands or a higher apparent molecular weight.
  • ELISA applications may be optimized using recombinant CD44 protein as a standard, and the antibody’s reactivity with the immunogen region (amino acids 346–385) should be validated in the relevant sample system.
  • Since the antibody cross-reacts with mouse CD44, it can be considered for rodent model studies; however, validate specificity in the target tissue or cell line.
  • Secreted forms of CD44 may be present in serum or cell culture supernatants; adjust sample preparation accordingly if detecting soluble CD44.
