Biological Background: CD44 Function and Localization

• CD44 is a single-pass type I membrane protein that serves as the primary receptor for hyaluronan (HA) and also interacts with collagen, growth factors, cytokines, and proteases, enabling cells to sense and respond to tissue microenvironment changes. • In immunohistochemistry and immunofluorescence, validate specificity using CD44 knockdown or knockout control cell lines/tissues to confirm staining patterns.
• For flow cytometry, optimize antibody titration and include isotype controls to accurately quantify CD44 expression on lymphocyte subsets, carcinoma cells, or other relevant populations.
• Given the shedding of CD44 ectodomain, sample preparation for ELISA-based assays should consider whether to detect total soluble CD44 or specific isoforms; include appropriate standards and blocking steps.
