Biological Background: CD44 Function and Localization
- CD44 is a single-pass type I membrane protein that serves as the primary receptor for hyaluronan (HA) and also interacts with collagen, growth factors, cytokines, and proteases, enabling cells to sense and respond to tissue microenvironment changes. - CD44 mediates lymphocyte activation, homing, and recirculation, as well as hematopoiesis and inflammatory responses to bacterial infection. Related references: PMID:7528188
- CD44 intracellular signaling involves the assembly of protein complexes with receptor kinases and membrane proteases, leading to activation of PKN2, RAC1, RHOA, and phospholipase C, which promote cytoskeletal reorganization and calcium mobilization. - The protein localizes primarily to the plasma membrane and microvilli, but a soluble ectodomain can be shed into the extracellular space.

Experimental Guidance and Technical Tips
- The immunogen encompasses the N-terminal hyaluronan-binding domain (aa 20–178), which is conserved and surface-exposed; this antibody may therefore detect multiple CD44 isoforms and native conformations in applications such as WB and IF/ICC.
- For Western blotting, the predicted molecular weight is ~82 kDa, but extensive glycosylation can cause a diffuse or ladder-like banding pattern; consider pretreatment with PNGase F or other deglycosylases to confirm specificity.
- In immunohistochemistry and immunofluorescence, validate specificity using CD44 knockdown or knockout control cell lines/tissues to confirm staining patterns.
- For flow cytometry, optimize antibody titration and include isotype controls to accurately quantify CD44 expression on lymphocyte subsets, carcinoma cells, or other relevant populations.
- Given the shedding of CD44 ectodomain, sample preparation for ELISA-based assays should consider whether to detect total soluble CD44 or specific isoforms; include appropriate standards and blocking steps.
