Biological Background: CD43 Function and Localization
- CD43 (Leukosialin, GPL115, GALGP, sialophorin) is the predominant cell surface sialoglycoprotein on leukocytes, encoded by the SPN gene. It is heavily glycosylated and plays critical roles in T-cell regulation.
- It regulates multiple T-cell functions including activation, proliferation, differentiation, trafficking, and migration. It positively regulates T-cell trafficking to lymph nodes via association with ERM proteins (EZR, RDX, MSN).
- Negatively regulates Th2 cell differentiation and promotes Th1 lineage commitment by enhancing IFN-gamma expression during TCR activation. Related references: PMID:18036228
- Prepares T-cells for cytokine sensing by inducing expression of cytokine receptors IFNGR and IL4R, promoting their signaling, and mediating clustering of IFNGR with TCR. Related references: PMID:24328034
- Acts as a major E-selectin ligand on Th17 cells, facilitating their rolling on activated vasculature and recruitment during inflammation. It mediates Th17 cell adhesion to E-selectin but not Th1 cell adhesion.
- Protects cells from apoptotic signals, thereby promoting cell survival.
- Subcellularly, CD43 localizes to the plasma membrane, with enrichment in cell projections such as microvilli and uropodium on migrating cells. It is also detected in the nucleus at PML bodies.
- Specifically expressed on thymocytes, T-lymphocytes, neutrophils, plasma cells, and myelomas.
Experimental Guidance and Technical Tips
- The immunogen is an N-terminal peptide (aa1-100) of human CD43, targeting the extracellular domain. This design may favor recognition of the native protein in immunohistochemistry.
- For IHC-P, consider using antigen retrieval buffers suitable for heavily glycosylated proteins. Validate staining on human lymphoid tissues (e.g., tonsil, lymph node) where CD43 is abundant.
- In ELISA applications, this antibody can be evaluated as a capture or detection reagent. Titrate antibody concentration and optimize blocking to minimize background from serum glycoproteins.
- Since CD43 is exclusively expressed on leukocytes, use appropriate negative controls (e.g., non-hematopoietic tissues) to confirm specificity.
