Biological Background: CD40L Function and Localization
- CD40L functions as a crucial cytokine ligand for CD40 (TNFRSF5) on antigen-presenting cells, essential for T-cell-dependent humoral immunity (PubMed:1280226, PubMed:31331973). Related references: PMID:1280226 PMID:31331973
- It provides costimulatory signals to T-cells, enhancing proliferation and the production of cytokines such as IL-4 and IL-10 upon TCR/CD3 ligation and CD28 costimulation (PubMed:8617933). Related references: PMID:8617933
- CD40L cross-linking triggers the NF-kappa-B pathway and activation of kinases MAPK8 and PAK2, and induces tyrosine phosphorylation of CD28 isoform 3 in T-cells (PubMed:15067037). Related references: PMID:15067037
- It mediates B-cell proliferation, immunoglobulin class switching, and IgE production in the presence of IL-4, highlighting its role in B-cell activation.
- CD40L is also an integrin ligand for ITGA5:ITGB1 and ITGAV:ITGB3, working in concert with CD40 to regulate signaling outcomes such as B-cell activation and anti-apoptotic responses.
- The protein is localized at the cell membrane and cell surface, and can be cleaved to produce a secreted form; it is specifically expressed on activated CD4+ T-lymphocytes.
- With a calculated molecular mass of ~29.3 kDa, glycosylation increases the observed size to around 33–39 kDa in its full-length active form.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 113–261, covering the extracellular TNF homology domain critical for receptor binding; this design may confer reactivity to both membrane-bound and soluble forms of CD40L.
- For Western blot (WB), the predicted molecular weight of unglycosylated CD40L is ~29 kDa, but glycosylated forms may appear around 33–39 kDa; verify band specificity using activated T-cell lysates or recombinant protein controls.
- For ELISA applications, consider using the antibody as a capture or detection reagent, paired with a validated CD40L matched antibody pair, and optimize coating with recombinant CD40L protein or cell-based assays.
- Cross-reactivity has been reported with human and mouse CD40L; validate in the selected species and assay system before use.
