Biological Background: CD4 Function and Localization
- CD4 is an integral membrane glycoprotein and member of the immunoglobulin superfamily, serving as a coreceptor for MHC class II molecules.
- In T-helper cells, CD4 interacts with the T-cell receptor (TCR) and MHC class II on antigen-presenting cells, recruiting the Src kinase LCK to initiate intracellular signaling cascades leading to lymphokine production, motility, adhesion, and T-cell activation.
- CD4 plays a critical role in thymic development of T-helper cells and triggers the differentiation of monocytes into mature macrophages.
- Expressed on the surface of T-helper cells, as well as on macrophages, dendritic cells, and NK cells, CD4 is a hallmark marker for T-helper cell identification.
- The protein undergoes post-translational modifications including glycosylation, phosphorylation, and palmitoylation, which regulate its membrane localization and function.
- CD4 also participates in TCR/LCK-independent pathways in non-T cells such as macrophages and NK cells, influencing differentiation, cytokine expression, and cell migration.
- Alternative splicing of the Cd4 gene generates multiple isoforms, contributing to functional diversity.
- As a key player in adaptive immunity, CD4 is essential for immune responses against pathogens and is a primary target in immunological research and diagnostic applications.
Experimental Guidance and Technical Tips
- For Western blot, the predicted molecular weight is ~51 kDa; ensure proper sample preparation and use appropriate loading controls.
- For immunohistochemistry on paraffin-embedded tissues (IHC-P), antigen retrieval methods should be optimized; consider testing with known CD4-expressing tissues such as lymph node or spleen.
- For immunofluorescence (IF-P), fixation and permeabilization conditions may affect staining; validate in your cell type of interest.
- For ELISA, this antibody may be used as a capture or detection reagent; pair with appropriate clones for sandwich ELISA development.
- The antibody is raised against recombinant CD4 (P01730) and cross-reacts with human, mouse, and cynomolgus monkey CD4; verify specificity in your assay system.
- Store according to supplier recommendations and avoid repeated freeze-thaw cycles.
