Biological Background: CD4 Function and Localization

  • CD4 is an integral membrane glycoprotein that serves as a coreceptor for MHC class II molecules, playing a central role in T-cell activation and immune responses.
  • Upon TCR engagement, CD4 recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling cascades that promote lymphokine production, adhesion, and T-helper cell differentiation.
  • CD4 is a primary receptor for HIV-1, facilitating viral entry; it is also a receptor for human herpesvirus 7 (HHV-7). Related references: PMID:12089508, PMID:16331979, PMID:2214026, PMID:17346169, PMID:7909607
  • In the thymus, CD4 participates in T-helper cell development, and it also triggers monocyte-to-macrophage differentiation.
  • CD4 is highly expressed on T-helper cells and is also present on macrophages, dendritic cells, and NK cells.
  • The protein is localized to the cell membrane and contains extracellular immunoglobulin-like domains, a transmembrane helix, and a cytoplasmic tail subject to palmitoylation and phosphorylation.

Experimental Guidance and Technical Tips

  • Because the conjugate is similar in spectral properties to FITC/Alexa Fluor 488, it is compatible with standard 488 nm laser lines for flow cytometry.
  • Given that the immunogen spans the extracellular domain (aa 26–207), the antibody is likely to recognize an extracellular epitope, making it suitable for live-cell staining without permeabilization.
  • Consider titrating the antibody to determine the optimal staining concentration that provides the best signal-to-noise ratio for your specific sample type.
  • Include appropriate positive and negative controls, such as human peripheral blood mononuclear cells (PBMCs) or known CD4+ and CD4- cell lines, to confirm specificity.
  • Avoid prolonged exposure to light to maintain fluorophore stability; store the antibody at 2–8 °C protected from light for short-term use, and aliquot and freeze at ‑20 °C for long-term storage.
  • As a monoclonal antibody, it offers consistent lot-to-lot performance and minimal background.
