Biological Background: CD3H (CD247) Function and Localization

  • CD3H, also known as CD247 or T-cell receptor T3 zeta chain, is the zeta subunit of the TCR-CD3 complex, essential for adaptive immune response and T-cell signal transduction.
  • The cytoplasmic domain contains immunoreceptor tyrosine-based activation motifs (ITAMs) that become phosphorylated by Src family kinases LCK and FYN upon TCR engagement, initiating downstream signaling. Related references: PMID:1384049, PMID:1385158, PMID:2470098
  • Phosphorylated ITAMs serve as docking sites for the protein kinase ZAP70, leading to ZAP70 phosphorylation and conversion into a catalytically active enzyme, a critical step in T-cell activation. Related references: PMID:7509083
  • CD247 plays an important role in intrathymic T-cell differentiation, influencing T-cell development in the thymus.
  • It also participates in activity-dependent synapse formation of retinal ganglion cells (RGCs) in both the retina and dorsal lateral geniculate nucleus (dLGN) (By similarity).
  • Subcellularly, CD247 localizes to the cell membrane.
  • CD247 is expressed in normal lymphoid tissue and in peripheral blood mononuclear cells (PBMCs). Related references: PMID:11722641
  • Keywords: Adaptive immunity, alternative splicing, host-virus interaction, phosphoprotein, transmembrane, reference proteome.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 50–164) covers the intracellular ITAM-containing region; thus, the antibody targets the cytoplasmic domain of CD247.
  • For WB, a band around 19 kDa is expected under reducing conditions; using Jurkat T-cell lysate as a positive control is advisable.
  • In IHC-P, antigen retrieval and permeabilization may be necessary to expose the intracellular epitope.
  • For ELISA, the antibody can be used for detection; optimal concentrations should be determined in the user's assay system.
  • Cross-reactivity with mouse and rat CD247 makes it suitable for rodent models, but confirm sequence homology for other species.
