Biological Background: CD3H (CD247) Function and Localization
- CD3H, also known as CD247 or T-cell receptor T3 zeta chain, is the zeta subunit of the TCR-CD3 complex, essential for adaptive immune response and T-cell signal transduction.
- The cytoplasmic domain contains immunoreceptor tyrosine-based activation motifs (ITAMs) that become phosphorylated by Src family kinases LCK and FYN upon TCR engagement, initiating downstream signaling. Related references: PMID:1384049, PMID:1385158, PMID:2470098
- Phosphorylated ITAMs serve as docking sites for the protein kinase ZAP70, leading to ZAP70 phosphorylation and conversion into a catalytically active enzyme, a critical step in T-cell activation. Related references: PMID:7509083
- CD247 plays an important role in intrathymic T-cell differentiation, influencing T-cell development in the thymus.

- In IHC-P, antigen retrieval and permeabilization may be necessary to expose the intracellular epitope.
