Biological Background: CD3E Function and Localization

  • CD3E is the epsilon chain of the T-cell surface glycoprotein CD3, which forms the TCR-CD3 complex on T lymphocytes and plays an essential role in adaptive immune responses. Related references: PMID:15294938, PMID:15546002, PMID:2470098
  • The CD3 chains (CD3D, CD3E, CD3G, CD247) contain immunoreceptor tyrosine-based activation motifs (ITAMs) in their cytoplasmic domains. Upon TCR engagement by antigen-MHC, these ITAMs are phosphorylated by Src kinases LCK and FYN, creating docking sites for ZAP70 and triggering downstream signaling cascades.   • Beyond signal transduction, CD3E is essential for proper T-cell development in the thymus (By similarity).
