Biological Background: CD3D Function and Localization
- CD3D encodes the T-cell surface glycoprotein CD3 delta chain, also known as the T-cell receptor T3 delta chain. It is a single-pass type I membrane protein and a key component of the TCR-CD3 complex.
- As part of the TCR-CD3 complex, CD3D is essential for adaptive immunity. Upon antigen recognition by the TCR, the CD3 chains transmit activation signals across the cell membrane via their immunoreceptor tyrosine-based activation motifs (ITAMs). - CD3D plays a critical role in thymocyte differentiation by ensuring correct TCR-CD3 complex assembly and surface expression. - In IHC-P, optimize antigen retrieval conditions and validate staining with known T-cell–rich tissues (e.g., tonsil, lymph node).
- For IF/ICC, permeabilization is required to detect CD3D, and co-staining with CD4/CD8 can help confirm T-cell specificity. Validate the protocol in your sample system.
- In ELISA, use the antibody as capture or detection reagent after confirming its performance with recombinant CD3D or cell lysates; cross-reactivity with rat CD3D allows multi-species assay design.
