Biological Background: CD3D Function and Localization
- CD3D encodes the delta chain of the T-cell surface glycoprotein CD3, a core component of the TCR-CD3 complex present on T lymphocytes. It is essential for adaptive immune response: upon TCR engagement by antigen-presenting cells, CD3 chains transmit signals across the cell membrane via their immunoreceptor tyrosine-based activation motifs (ITAMs), leading to activation of downstream signaling pathways.

- The immunogen is a synthetic peptide covering amino acids 72–171 of human CD3D; this peptide-based design may favor recognition of denatured protein in Western blot (WB) applications. Consider using low molecular weight markers and optimal transfer conditions for the 19 kDa target.
- For immunofluorescence (IF-P) and flow cytometry (FC), ensure proper permeabilization and fixation protocols depending on whether the epitope is intracellular or surface-accessible. Validate the antibody in relevant T-cell lines (e.g., Jurkat) or primary human PBMCs.
- In ELISA, pair with a capture/detection antibody validated for the CD3D target to build a sensitive sandwich assay. Cross-reactivity with other species has not been tested; use human samples only.
