Biological Background: CD39/ENTPD1 Function and Localization
- CD39/ENTPD1 (ectonucleoside triphosphate diphosphohydrolase 1) is a transmembrane enzyme that sequentially hydrolyzes extracellular nucleoside triphosphates (NTPs) to diphosphates (NDPs) and then to monophosphates (NMPs), thus controlling the availability of signaling nucleotides. Related references: PMID:8529670 PMID:8626624 PMID:8955160
- By degrading pro-inflammatory ATP and platelet-activating ADP, CD39 inhibits platelet aggregation and promotes vascular homeostasis, underscoring its role in thromboregulation. Related references: PMID:8955160 PMID:8996251
- CD39 is highly expressed on activated lymphoid cells and endothelial tissues, with additional expression in placenta, lung, skeletal muscle, and kidney.
- The protein localizes to the plasma membrane and caveolae, consistent with its function in regulating extracellular nucleotide pools.
- It contains conserved ATP/GTP-binding domains, disulfide bonds, and multiple glycosylation sites, classifying it as a calcium- and magnesium-dependent hydrolase.
- Mutations in ENTPD1 are linked to hereditary spastic paraplegia type 64 (SPG64), highlighting its importance beyond the vasculature.
- Alternative splicing of ENTPD1 produces isoforms that may affect enzymatic activity or localization.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 255–442 of the extracellular domain, making the antibody suitable for surface staining of live cells in flow cytometry without permeabilization.
- For ELISA, consider pairing with a different clone or using it as a capture antibody for soluble CD39, and validate with appropriate biological samples.
- Although validated for human reactivity, cross-reactivity with other species should be confirmed experimentally if needed.
