Biological Background: CD38 Function and Localization
- CD38 is a multifunctional transmembrane glycoprotein also known as ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1, with aliases including I-19, ADPRC 1, and NIM-R5 antigen.
- It synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion, and also produces nicotinate-adenine dinucleotide phosphate (NAADP+) from 2'-phospho-cADPR and nicotinic acid. Related references: PMID:11829748
- CD38 functions as a receptor binding CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and migration across endothelial barriers.
- In dendritic cells, CD38 regulates crucial mature-stage functions such as CCL21-driven migration, survival, and Th1-polarizing activity.
- Subcellularly, CD38 is a membrane protein with a signal-anchor and transmembrane helix; it is a glycoprotein with disulfide bonds and possesses both hydrolase and transferase enzymatic activities.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 45–304, a large extracellular region; this design may yield antibodies recognizing native CD38 in mouse samples.
- For Western blotting, a band at approximately 34 kDa is anticipated; validate using mouse tissue or cell lysates known to express CD38.
- In IF/ICC, membrane staining should be observed; appropriate permeabilization may enhance detection.
- Flow cytometry can be performed on mouse lymphocyte populations; titrate the antibody to optimize the signal.
- ELISA protocols can be developed using either direct antigen coating or sandwich formats; consider pairing with other CD38-specific reagents.
